GID complex regulates the differentiation of neural stem cells by destabilizing TET2

摘要: Brain development requires a delicate balance between self-renewal and differentiation in neural stem cells (NSC), which rely on the precise regulation of gene expression. Ten-eleven translocation 2 (TET2) modulates gene expression by the hydroxymethylation of 5-methylcytosine in DNA as an important epigenetic factor and participates in the neuronal differentiation. Yet, the regulation of TET2 in the process of neuronal differentiation remains unknown. Here, the protein level of TET2 was reduced by the ubiquitin-proteasome pathway during NSC differentiation, in contrast to mRNA level. We identified that TET2 physically interacts with the core subunits of the glucose-induced degradation-deficient (GID) ubiquitin ligase complex, an evolutionarily conserved ubiquitin ligase complex and is ubiquitinated by itself. The protein levels of GID complex subunits increased reciprocally with TET2 level upon NSC differentiation. The silencing of the core subunits of the GID complex, including WDR26 and ARMC8, attenuated the ubiquitination and degradation of TET2, increased the global 5-hydroxymethylcytosine levels, and promoted the differentiation of the NSC. TET2 level increased in the brain of the Wdr26+/− mice. Our results illustrated that the GID complex negatively regulates TET2 protein stability, further modulates NSC differentiation, and represents a novel regulatory mechanism involved in brain development.

Monocyte subsets and their differentiation tendency after burn injury

摘要:

Monocytes are critical effectors and regulators of immune response. Studying the nomenclature of monocyte subsets may be beneficial for understanding the complex function of monocytes in steady and inflammatory states. A monocyte has the potential to differentiate into dendritic cells or macrophages, and this behavior significantly changes in severely burned patients and mice. The findings in the present study may help enhance understanding on the perturbation of the immune system after severe burn injury.

Protective roles of heat stress on the neurons in hippocampal CA1 region of mice

摘要: The effects of heat stress on the neurons in hippocampal CA1 region of brain ischemia/reperfusion were explored. The mice were pretreated with heat stress followed by ischemia/reperfusion by clipping bilateral cervical commo n arteries for 7 min. Mice were divided randomly into four groups as follows: (1) normal control group; (2) heat stress pretreated subsequent to ischemia/reperfusion group (HS/IR); (3) ischemia/reperfusion group (IR); and (4) heat stress group (HS). Animals in the last three groups were subdivided into three subgroups: 1 d, 4 d, 14 d respectively. The Morris water maze was used to test the ability of learning and memorizing, Nissl staining was used to count the average number of survived neurons in hippocampal CA1 region, and immunohistochemistry combined with image analysis system to detect the changes of Microtubule associated protein 2 (MAP-2) expression. The results showed that mice in IR group exhibited increased escape latency when compared with that of normal, HS and HS/IR groups (<0.01), and the mice in IR group adopted an inefficient search strategy, major in circling and restricted searching manners. Nissl staining results showed a significant reduction in the number of pyramidal neurons in hippocampal CA1 regions in HS/IR and IR groups, with a decrease in IR group (<0.01). Compared with normal group, the expression of MAP-2 in hippocampal CA1 region obviously decreased in IR group (<0.05). The present results indicate that heat stress pretreatment can improve the spatial learning and memorizing function through protection to hippocampal neurons.

摘要: This study aimed to investigate the protective effect of nicotine on dopaminergic neurons and its mechanisms in mice with Parkinson disease (PD) induced by 1-methyl-4-phenyl-1,2,3, 6-tetrahydropyridine (MPTP). C57BL/6J mice were injected with MPTP for 8 days to establish a PD model. Nicotine was given for 10 days in the nicotine therapeutic group. Animals were examined behaviorally with the pole test and traction test. Tyrosine hydroxylase (TH) and γ-aminobutyric acid (GABA) were determined by using the immunocytochemistry (ICC) method. The ultrastructural changes of the caudate nucleus (CN) were observed under electron microscopy. The results showed that pretreatment with nicotine could improve the dyskinesia of PD mice markedly. Simultaneously, TH-positive ( < 0.01) neurons and GABA-positive ( < 0.05) neurons in the nicotine therapeutic group were significantly more than those in the model group. The ultrastructural injury of the nicotine therapeutic group was also ameliorated. Nicotine has protective effects on the γ-aminobutyric acid neurons and dopaminergic neurons in the MPTP-treated mice.

Abivertinib inhibits megakaryocyte differentiation and platelet biogenesis

摘要: Abivertinib, a third-generation tyrosine kinase inhibitor, is originally designed to target epidermal growth factor receptor (EGFR)-activating mutations. Previous studies have shown that abivertinib has promising antitumor activity and a well-tolerated safety profile in patients with non-small-cell lung cancer. However, abivertinib also exhibited high inhibitory activity against Bruton’s tyrosine kinase and Janus kinase 3. Given that these kinases play some roles in the progression of megakaryopoiesis, we speculate that abivertinib can affect megakaryocyte (MK) differentiation and platelet biogenesis. We treated cord blood CD34+ hematopoietic stem cells, Meg-01 cells, and C57BL/6 mice with abivertinib and observed megakaryopoiesis to determine the biological effect of abivertinib on MK differentiation and platelet biogenesis. Our in vitro results showed that abivertinib impaired the CFU-MK formation, proliferation of CD34+ HSC-derived MK progenitor cells, and differentiation and functions of MKs and inhibited Meg-01-derived MK differentiation. These results suggested that megakaryopoiesis was inhibited by abivertinib. We also demonstrated in vivo that abivertinib decreased the number of MKs in bone marrow and platelet counts in mice, which suggested that thrombopoiesis was also inhibited. Thus, these preclinical data collectively suggested that abivertinib could inhibit MK differentiation and platelet biogenesis and might be an agent for thrombocythemia.

摘要: The secondary laticifer, a specific tissue in the secondary phloem of rubber tree, is differentiated from the vascular cambia. The number of the secondary laticifer in the trunk bark of rubber tree is positively correlated with rubber yield. Although jasmonates have been demonstrated to be crucial in the regulation of secondary laticifer differentiation, the mechanism for the jasmonate-induced secondary laticifer differentiation remains to be elucidated. By using an experimental morphological technique, the present study revealed that trichostatin A (TSA), an inhibitor of histone deacetylation, could induce the secondary laticifer differentiation in a concentration-dependent manner. The results suggest that histone acetylation is essential for the secondary laticifer differentiation in rubber tree.

Spontaneous firing properties of rat medial vestibular nucleus neurons in brain slices by infrared visual

摘要: Domestic application of infrared patch clamp techniques on brain slices is limited. The key of the technique is to prepare high-quality brain slices. The present paper describes the preparation procedure of brainstem slices and the spontaneous firing properties of rat medial vestibular nucleus (MVN) neurons. By infrared differential interference contrast technique, neurons of rat MVN were visualized directly at the depth of 50–100 ?m underneath the surface of slices. Firing activities of MVN neurons were recorded by the whole-cell patch clamp technique in artificial cerebrospinal fluid (ACSF) and low Ca - high Mg fluid. The firing mode was more irregular and depressive in low Ca - high Mg fluid than in ACSF. According to the averaged waveform of action potentials, cells were classified as the neurons with monophasic after-hyperpolarization potential (AHP), and the neurons with biphasic AHP. The resting membrane potential (RMP), input resistance (Rin) and membrane capacitance (Cm) of neurons were recorded and compared between groups. With infrared videomicroscopy, patch clamp recordings could be made under direct observation in freshly prepared brainstem slices. The discharge activities of MVN neurons were spontaneous and the firing mode was modulated by extracellular calcium concentration. The basic membrane properties of two types of neurons were not significantly different, while the differences in waveform might play a role in the segregation between tonic and kinetic cells.

coupling relationship of industrial agglomeration and low-carbon economy considering spatiotemporal differentiation

摘要: Although China’s construction machinery thrives to meet the needs of construction, a number of challenges still remain to be overcome, such as lack of thorough knowledge of regional disparities and several limitations in terms of carbon emissions and economic development. Meanwhile, a low-carbon economy was proposed and implemented in China. This research aims to investigate the differences in industrial agglomeration of construction machineries and further explore the relationship between industrial agglomeration and low-carbon economy. On this basis, spatiotemporal analysis was performed to evaluate the levels of industrial agglomeration in different regions based on the situations of China’s construction machinery industry. Furthermore, this study explored the interaction between industrial agglomeration and low-carbon economy utilizing the coupling coordination analysis method. Results showed that the coupling coordination of the two subsystems was extremely unbalanced in 2006, and it maintained an increasing trend, reaching a relatively high level in 2018. Finally, suggestions, such as establishing a policy guarantee system and implementing variable policies in different regions, were proposed to provide guidelines for the government decision-making and promote the sustainable development of China’s construction machinery industry.

Repression of CDKN2C caused by PML/RARα binding promotes the proliferation and differentiation block

Inappropriate cell proliferation during oncogenesis is often accompanied by inactivation of components involved in the cell cycle machinery. Here, we report that cyclin-dependent kinase inhibitor 2C (CDKN2C) as a member of the cyclin-dependent kinase inhibitors is a target of the PML/RARα oncofusion protein in leukemogenesis of acute promyelocytic leukemia (APL). We found that CDKN2C was markedly downregulated in APL blasts compared with normal promyelocytes. Chromatin immunoprecipitation combined with quantitative polymerase chain reaction demonstrated that PML/RARα directly bound to the CDKN2C promoter in the APL patient-derived cell line NB4. Luciferase assays indicated that PML/RARα inhibited the CDKN2C promoter activity in a dose-dependent manner. Furthermore, all-trans retinoic acid treatment induced CDKN2C expression by releasing the PML/RARα binding on chromatin in NB4 cells. Functional studies showed that ectopic expression of CDKN2C induced a cell cycle arrest at the G0/G1 phase and a partial differentiation in NB4 cells. Finally, the transcriptional regulation of CDKN2C was validated in primary APL patient samples. Collectively, this study highlights the importance of CDKN2C inactivation in the abnormal cell cycle progression and differentiation block of APL cells and may provide new insights into the study of pathogenesis and targeted therapy of APL.

Cannabidiol prevents depressive-like behaviors through the modulation of neural stem cell differentiation

摘要: Chronic stress impairs radial neural stem cell (rNSC) differentiation and adult hippocampal neurogenesis (AHN), whereas promoting AHN can increase stress resilience against depression. Therefore, investigating the mechanism of neural differentiation and AHN is of great importance for developing antidepressant drugs. The nonpsychoactive phytocannabinoid cannabidiol (CBD) has been shown to be effective against depression. However, whether CBD can modulate rNSC differentiation and hippocampal neurogenesis is unknown. Here, by using the chronic restraint stress (CRS) mouse model, we showed that hippocampal rNSCs mostly differentiated into astrocytes under stress conditions. Moreover, transcriptome analysis revealed that the FoxO signaling pathway was involved in the regulation of this process. The administration of CBD rescued depressive-like symptoms in CRS mice and prevented rNSCs overactivation and differentiation into astrocyte, which was partly mediated by the modulation of the FoxO signaling pathway. These results revealed a previously unknown neural mechanism for neural differentiation and AHN in depression and provided mechanistic insights into the antidepressive effects of CBD.

Industrial eco-efficiency and its spatial-temporal differentiation in China

摘要: The aim of this paper is to study the spatial-temporal differentiation of industrial eco-efficiency in China. Using methods based on the data envelopment analysis (DEA) model and exploratory spatial data analysis (ESDA) and data from 1985, 1995, 2005, and 2008 of 30 provinces in China, the spatial-temporal pattern changes in industrial eco-efficiency are discussed. The results show that: first, the patterns of industrial eco-efficiency are dominated by clustering of relatively low efficiency provinces; second, spatial relationships between the industrial eco-efficiencies of different provinces changed slightly throughout the period and the provinces persistently exhibit spatial concentration of relatively low industrial eco-efficiency; finally, there is an obvious trend in the polarization of industrial eco-efficiency, i.e., the higher level spatial units are concentrated in eastern China, and the lower level spatial units are mainly in western and central China.

艾滋病是一种获得性免疫缺陷综合征,中医学的观点则是正气严重不足。正气依靠精神的支撑,而精神来源于饮食气味及其精微物质 ,精微物质的转化主要靠脏腑功能的正常运化。脏腑功能活动是生命运动的根本。五脏各有所主,各司其职,并且相互和合与平衡,只有当五 脏功能旺盛,并且相互和合时,人体的生命力才会强大,机体“正气”充足,才会有免疫力。在五脏之中,肝、脾、肾三脏功能要保持平衡, 其他各脏功能才会自然趋向平衡与协调,人体才会健康长寿,这就是“三维守恒”法则。

Resveratrol promotes the survival and neuronal differentiation of hypoxia-conditioned neuronal progenitor

摘要: Hypoxia conditioning could increase the survival of transplanted neuronal progenitor cells (NPCs) in rats with cerebral ischemia but could also hinder neuronal differentiation partly by suppressing mitochondrial metabolism. In this work, the mitochondrial metabolism of hypoxia-conditioned NPCs (hcNPCs) was upregulated via the additional administration of resveratrol, an herbal compound, to resolve the limitation of hypoxia conditioning on neuronal differentiation. Resveratrol was first applied during the neuronal differentiation of hcNPCs and concurrently promoted the differentiation, synaptogenesis, and functional development of neurons derived from hcNPCs and restored the mitochondrial metabolism. Furthermore, this herbal compound was used as an adjuvant during hcNPC transplantation in a photothrombotic stroke rat model. Resveratrol promoted neuronal differentiation and increased the long-term survival of transplanted hcNPCs. 18-fluorine fluorodeoxyglucose positron emission tomography and rotarod test showed that resveratrol and hcNPC transplantation synergistically improved the neurological and metabolic recovery of stroke rats. In conclusion, resveratrol promoted the neuronal differentiation and therapeutic efficiency of hcNPCs in stroke rats via restoring mitochondrial metabolism. This work suggested a novel approach to promote the clinical translation of NPC transplantation therapy.
